The Zymoprep Yeast Plasmid Miniprep II is a simple and efficient yeast plasmid miniprep that is based on the E. coli alkaline lysis method but using Zymolyase as the first solution. The kit provides all the necessary reagents for plasmid isolation from S. cerevisiae, C. albicans and S. pombe, and any fungi whose cell walls are susceptible to yeast lytic enzyme lysis. The procedure is simple and efficient, with no need for glass beads or phenol. The Zymoprep II recovers about 5 fold more plasmid DNA compared to Zymoprep I and eliminates the isopropanol precipitation step. Reliably recover plasmid DNA from yeast colonies, patches on plates, or as liquid cultures. The system is ideal for low-copy number and hard-to-isolate plasmids. Eluted plasmid DNA can be used directly for E. Coli transformation, PCR, and Southern blot analysis.

Ethanol must be added to the concentrate prior to use. Instructions for each DNA Wash Buffer (concentrate) size is listed on the bottle and within the Zymo Research protocols it is used in.

The E.Z.N.A. FastFilter Plasmid Maxi Kit combines the convenience of spin columns with the speed of syringe filters to ease large-scale plasmid preparations. The system utilizes Lysate Clearance Filter Syringes instead of centrifugation to rapidly clear bacterial lysates post-alkaline lysis and follows a simple “bind-wash-elute” procedure to deliver high-quality plasmid DNA. The yields vary according to plasmid copy number, E. coli strain, and growth conditions. 50-200 mL of bacterial cultures in LB medium typically produces 0.6-1.2 mg of high copy number plasmid DNA. Up to 500 mL of culture may be processed when working with low copy number plasmid. The system uses centrifugation or vacuum technology for plasmid purification and eliminates the time-consuming gravity-flow columns that require alcohol precipitation.
